研究專長

免疫學、自體免疫疾病之機轉及治療之研究

本實驗室主要研究領域在自體免疫疾病致病機轉的探討及樹突狀細胞之研究。樹突狀細 胞是目前已知具最佳免疫調控能力之抗原呈現細胞(antigen presentation cell)。

探討第三誘餌蛋白質對自體免疫糖連病小鼠骨髓樹突狀細胞分化之調控。

「第三誘餌蛋白質(decoy receptor 3)」對自體免疫糖連病小鼠骨髓樹突狀細胞(bone marrow-derived dendritic cell)分化之調控;並以此方法發展出新型治療策略。樹突狀細胞是目前已知具最佳免疫調控能力之抗原呈現細胞(antigen presentation cell),經三誘餌蛋白質處理過後之樹突狀細胞具有誘導T細胞走向第二型輔助性免疫反應(T-helper 2 immune response)之特性,恰好可以阻抗因第一型輔助性免疫反應(T-helper 1 immune response)所主導之自體免疫糖連病。本研究以昆蟲細胞系統(Sf21-bacculoexpressing system)生產並純化高品質第三誘餌蛋白質,並以此T細胞走向第二型輔助性免疫反應,且於細胞移植實驗(dendritic cell transfer experiment)中證實其免疫保護效果。另外更以先進之蛋白質體技術(proteeomic analysis)分析此誘餌蛋白對樹突狀細胞分化調控過程中各類基因細微之變化,此部分結果發表於 Journal of Leukocyte Biology 2004

延續過去的研究,未來亦將應用於另一種自體免疫疾病:多發性硬化症(multiple sclerosis)之研究。
